This video shows luminescence arising the frq promoter driving luciferase in frq luc-I Neurospora strains, bottom view on a race tube at 25°C over about 10 circadian cycles. Race tube medium: (0.1 % glucose, 0.17 % arginine, 1X Vogel's, 50 ug/L biotin, 1.8 % agar, 50 uM luciferin). Image is taken every 30 min. Time is h in DD after transfer from light. Taken from Gooch VD, Mehra A, Larrondo LF, Fox J, Touroutoutoudis M, Loros JJ, Dunlap JC. Fully codon-optimized luciferase uncovers novel temperature characteristics of the Neurospora clock. This video shows Neurospora bearing a transgene in which the ccg-2 promoter drives codon-optimized luciferase. Luminescence arising from eas/ccg 2-luc-I Neurospora strains is false-colored and seen side view on a race tube at 25°C over 3 days. Race tube medium: (0 % glucose, 0.17 % arginine, 1X Vogel's, 50 ug/L biotin, 1.5 % agar, 42 uM luciferin). Colors represent intensity, white to red being brightest and blues dimmest. Images were taken every 30 min. Time is in units of d:h:m. Note the agar surface traversing approximately through the center of the images.
